Western blotting experiments were conducted to assess the effects of QTJD on the MAPK signaling pathway components including ERK1/2, p38, and JNK1/2. The study investigated how QTJD's therapeutic effects are mediated through butyrate-induced modulation of MAPK signaling. Results showed that QTJD treatment significantly inhibited the phosphorylation/activation of ERK1/2, p38, and JNK1/2 MAPK proteins, suggesting that QTJD exerts its anti-inflammatory effects by suppressing pro-inflammatory MAPK signaling cascades involved in macrophage activation and cytokine production.
PRIMARY OUTCOME
MAPK protein phosphorylation levels
EXPECTED OUTCOMES
QTJD inhibits MAPK pathway activation
SUCCESS CRITERIA
Significant reduction in MAPK protein phosphorylation
PROTOCOL
Western blotting analysis of ERK1/2, p38, and JNK1/2 phosphorylation following QTJD treatment
