🧫 Experiment Protocol
ExploratoryatherosclerosisTBK1mouse and human arteries with atherosclerotic plaquesproposed
Liquid chromatography-mass spectrometry, immunostaining, RNA sequencing, and Western blot analyses were performed on mouse and human arteries with atherosclerotic plaques to identify key mediators of endothelial-to-mesenchymal transition (EndMT) and atherogenesis. TBK1 was identified as one of the key mediators through these comprehensive proteomic and transcriptomic approaches. The study compared atherosclerotic arteries with controls to determine differential expression patterns of proteins involved in endothelial dysfunction and plaque formation.
PRIMARY OUTCOME
identification of TBK1 as key mediator of EndMT
EXPECTED OUTCOMES
identification of proteins driving endothelial dysfunction and atherosclerosis
SUCCESS CRITERIA
differential expression of key proteins in atherosclerotic vs control arteries
PROTOCOL
liquid chromatography-mass spectrometry, immunostaining, RNA sequencing, Western blot analysis
