Multiomics, spatial genomics, and quantitative neuropathology applied to BRAIN Initiative reference atlases. Donors (33 male, 51 female; mean age 88) were stratified using a disease pseudoprogression score based on neuropathology.
PRIMARY OUTCOME
Multimodal cell atlas analysis of 84 human AD donor brains identified two disease phases: an early phase with inflammatory microglia, reactive astrocytes, somatostatin+ inhibitory neuron loss, and remyelination by OPCs; and a later phase with exponential pathology increase, excitatory neuron loss, and Pvalb+/Vip+ inhibitory neuron subtype loss.
