Motor neurons exhibit unique metabolic dependencies on glycolysis and lipid metabolism that become dysregulated in ALS. Targeting PDH kinase or SGLT2 to shift toward glucose oxidation may restore energy homeostasis. However, causal linkage to ALS progression is weak, and SOD1 metabolic rescue is a poor predictor for sporadic ALS.
