Phase 2 RCT comparing GFAP modulator vs. placebo in N=300 early AD patients, primary endpoint: change from baseline on composite cognitive score at 18 months. Sample size powered at 80% for effect size=0.28, alpha=0.05.

Kill criteria
- GFAP-reactive astrocyte subtyping shows no association with regional vulnerability
- Astrocyte GFAP knockdown fails to alter disease trajectory in AD mouse model
- Astrocyte-targeted intervention does not modify neuronal survival outcomes
