Motor neurons affected by C9orf72 repeats typically have expanded GGGGCC hexanucleotide repeats (normally 2-24 repeats) that exceed 30 copies, triggering multiple pathogenic cascades. The selective vulnerability of these neurons despite ubiquitous C9orf72 expression suggests specific cellular vulnerabilities tied to motor neuron biology rather than simple gene dosage effects.
