The Horizontal Diagonal Band (HDB) is a basal forebrain cholinergic nucleus that plays crucial roles in olfactory processing, memory formation, and cortical arousal. As part of the septal diagonal complex, HDB cholinergic neurons provide extensive projections to the olfactory bulb, hippocampus, and entorhinal cortex, making them essential for odor-guided behavior, spatial memory, and the integration of sensory information with limbic circuits. Anatomy and Morphology
Location and Organization The diagonal band of Broca consists of two limbs:
Vertical limb (VDB) : Projects to the hippocampus
Horizontal limb (HDB) : Projects to the olfactory bulb and cortex
HDB neurons are distributed along the horizontal fiber bundle extending from the septal region toward the olfactory tubercle. The nucleus is topographically organized, with different subregions preferentially targeting different downstream structures.
Subdivisions
Ch1 : Most medial portion, primarily cholinergic
Ch2 : Intermediate region, mixed cholinergic/GABAergic
Ch3 : Lateral portion, projection to olfactory bulb
Connectivity
Olfactory bulb : Centrifugal feedback projections
Piriform cortex : Cortical feedback
Hippocampus : Via medial septum
Hypothalamus : Preoptic area, lateral hypothalamus
Brainstem : Raphe nuclei, locus coeruleus
Efferent Projections
Olfactory bulb : Dense cholinergic innervation of granule and mitral cells
Anterior olfactory nucleus : Secondary olfactory processing
Piriform cortex : Primary olfactory cortex
Entorhinal cortex : Layer II stellate cells
Hippocampus : Via lateral septum
Electrophysiology
Intrinsic Properties HDB cholinergic neurons display:
Resting membrane potential : -55 to -65 mV
Input resistance : 80-150 MΩ
Membrane time constant : 15-25 ms
Firing rate : 5-15 Hz spontaneous activity
Firing Patterns
Tonic firing : Steady-state activity during wakefulness
Burst firing : Synchronized bursts during odor sampling
Silent states : Reduced activity during slow-wave sleep
Cholinergic Modulation HDB cholinergic activity modulates target circuits by:
Increasing signal-to-noise ratio in olfactory processing
Enhancing memory consolidation
Modulating cortical plasticity
Molecular Signature
Cholinergic Markers
Choline acetyltransferase (ChAT) : Acetylcholine synthesis
Acetylcholinesterase (AChE) : Signal termination
Vesicular acetylcholine transporter (VAChT) : Vesicular packing
Receptor Expression
Muscarinic receptors : M1, M2, M3 subtypes on target neurons
Nicotinic receptors : α4β2, α7 on afferent terminals
Role in Neurodegenerative Diseases
Alzheimer's Disease HDB neurons are affected in AD but typically less severely than medial septum:
Moderate reduction in cholinergic neuron number
Decreased ChAT activity
Early olfactory dysfunction correlates with HDB pathology
Contributes to early smell identification deficits
Parkinson's Disease
Lewy bodies can form in HDB cholinergic neurons
Contributes to olfactory dysfunction (early PD symptom)
Interaction with limbic alpha-synuclein pathology
Other Disorders
Olfactory dysfunction : Schizophrenia, depression
Memory deficits : Age-related cognitive decline
Therapeutic Implications
Targeting HDB for Treatment
Cholinergic agonists : Direct muscarinic activators
Olfactory delivery : Nose-to-brain drug delivery to target HDB
Deep brain stimulation : Targeting diagonal band for memory
Olfactory training : Sensory stimulation to preserve function
