The raphe nuclei constitute the principal source of serotonergic innervation in the mammalian brain. These brainstem nuclei contain the cell bodies of serotonin-producing neurons that project to virtually all regions of the central nervous system. Serotonin (5-hydroxytryptamine, 5-HT) is a critical neuromodulator that influences mood, arousal, sleep, appetite, pain perception, and cognitive function. In neurodegenerative diseases such as [Alzheimer's disease](/diseases/alzheimers-disease) (AD) and [Parkinson's disease](/diseases/parkinsons-disease) (PD), raphe nuclei undergo significant degeneration, contributing to the non-motor symptoms that often precede motor manifestations.
