Overview

Sigma-1 Receptor Neurons are neurons expressing the σ1 receptor (σ1R), a unique intracellular chaperone protein that has garnered substantial attention in neurodegenerative disease research due to its pleiotropic protective effects. The σ1 receptor is a 223-amino acid protein localized primarily to the endoplasmic reticulum (ER) membrane, particularly at mitochondria-associated ER membranes (MAMs), where it serves as a dynamic scaffold that organizes signaling complexes and regulates multiple cellular processes including calcium homeostasis, mitochondrial function, protein folding, and cellular stress responses. These receptor neurons are abundantly expressed throughout the brain, with particularly high levels in the hippocampus, cerebral cortex, basal ganglia, cerebellum, and spinal cord, where they participate in modulating neurotransmission, synaptic plasticity, and neuronal survival. The σ1 receptor has emerged as a promising therapeutic target for Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis (ALS), and other neurodegenerative conditions due to its central role in maintaining cellular homeostasis under stress conditions.
