Introduction
TDP-43 proteinopathy [neurons](/entities/neurons) represent a defining pathological feature of amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration with TDP-43 pathology (FTLD-TDP). These neurodegenerative conditions share a common pathological hallmark: the aggregation of the TAR DNA-binding protein 43 (TDP-43) into cytoplasmic inclusions within neurons and glia [1](https://pubmed.ncbi.nlm.nih.gov/17023659/). TDP-43 is a 414-amino acid nuclear protein encoded by the TARDBP gene that functions in RNA processing, splicing, transport, and translation regulation. In affected neurons, TDP-43 is mislocalized from the nucleus to the cytoplasm, where it forms insoluble aggregates that are ubiquitinated and hyperphosphorylated [2](https://pubmed.ncbi.nlm.nih.gov/18600239/).
