Overview

Donepezil (brand name Aricept; also known as E2020) is a piperidine-based, selective, and reversible acetylcholinesterase (AChE) inhibitor used for the symptomatic treatment of [Alzheimer's disease](/diseases/alzheimers-disease) (AD) across all severity stages. Developed by Eisai Co., Ltd. and co-marketed with Pfizer, donepezil received FDA approval in 1996 for mild-to-moderate AD and remains the most widely prescribed AD medication worldwide. Its long half-life of approximately 70 hours permits convenient once-daily dosing, distinguishing it from other [cholinesterase inhibitors](/entities/cholinesterase-inhibitors) (ChEIs) such as [rivastigmine](/therapeutics/rivastigmine) and [galantamine](/therapeutics/galantamine) ([Sugimoto et al., 2002](https://doi.org/10.2174/1568026023394674)).
