Necroptosis

Introduction

Necroptosis is an important component in the neurobiology of neurodegenerative diseases. This page provides detailed information about its structure, function, and role in disease processes.

Overview

Necroptosis is a regulated form of programmed cell death that combines features of [apoptosis](/entities/apoptosis) (programmed execution) with necrotic morphology (cell swelling and membrane rupture). Unlike [apoptosis](/entities/apoptosis), necroptosis is caspase-independent and is mediated by the RIPK1–RIPK3–MLKL signaling axis. First identified as a "backup" death pathway when [caspases](/entities/caspases) are inhibited, necroptosis is now recognized as a physiologically significant process with critical roles in development, innate immunity, and disease pathogenesis. In the context of neurodegeneration, necroptosis has emerged as a major driver of neuronal loss and [neuroinflammation](/mechanisms/neuroinflammation) in [Alzheimer's disease](/diseases/alzheimers-disease), [Parkinson's disease](/diseases/parkinsons-disease), [amyotrophic lateral sclerosis](/diseases/amyotrophic-lateral-sclerosis), and [multiple sclerosis](/diseases/multiple-sclerosis) ([Bhardwaj et al., 2025](https://link.springer.com/article/10.1007/s10571-025-01601-w); [Bhargava et al., 2024](https://link.springer.com/article/10.1007/s00401-024-02747-5)).
