Overview

Simufilam (formerly known as SAV-101) is a small molecule drug candidate developed by [Cassava Sciences](/companies/cassava-sciences) for the treatment of [Alzheimer's disease](/diseases/alzheimers-disease) (AD). It represents a fundamentally different approach from traditional AD therapeutics, targeting the scaffolding protein filamin A (FLNA) rather than directly attacking [amyloid-beta](/proteins/amyloid-beta) plaques or [tau](/proteins/tau) neurofibrillary tangles[@martinezluaces2022].

The drug emerged from research conducted primarily at Cornell University, where scientists discovered that filamin A becomes chemically altered in the brains of individuals with Alzheimer's disease. This alteration affects how the protein interacts with the [amyloid precursor protein](/entities/app-protein) (APP), contributing to the pathological production of amyloid-beta[@cataldo2020].

Filamin A Biology and Alzheimer's Disease

Normal Filamin A Function

Filamin A is a large (280 kDa) actin-binding protein that serves as a critical scaffolding molecule in neurons and other cell types. It performs several essential functions[@popa2021]:
