Background and Rationale

Amyotrophic lateral sclerosis is a fatal neurodegenerative disease affecting both upper motor neurons (UMNs) in the cortex and lower motor neurons (LMNs) in the brainstem and spinal cord. The disease was historically viewed as a focal, stochastic process, but converging evidence from post-mortem neuropathology, in vivo neuroimaging, and molecular biology now supports a network-based propagation model — ALS begins in specific network epicenters and spreads along structurally and functionally connected neural pathways.
