AXIN1 (Axis Inhibition Protein 1) is a critical scaffolding protein that plays a central role in regulating the Wnt/beta-catenin signaling pathway. AXIN1 encodes a protein of approximately 862 amino acids that functions as a negative regulator of canonical Wnt/beta-catenin signaling. AXIN1 forms a complex with APC, [GSK-3beta](/entities/gsk3-beta), and CK1alpha (the destruction complex) that phosphorylates beta-catenin, targeting it for ubiquitination and proteasomal degradation.
Beyond its canonical role in Wnt signaling, AXIN1 participates in various cellular processes including the DNA damage response, JNK signaling, TGF-beta signaling, and microtubule regulation. Molecular Structure
The AXIN1 protein contains several functional domains:
