MFNG (Manic Fringe) encodes a Golgi-localized glycosyltransferase that modifies Notch receptors by adding N-acetylglucosamine (GlcNAc) to O-fucose residues on the extracellular domain. This post-translational modification specifically enhances DLL1-mediated Notch signaling while reducing JAG1-mediated activation[@gridley2023].
