Overview

NPAS3 (Neuronal PAS Domain Protein 3) encodes a brain-specific transcription factor belonging to the bHLH-PAS (basic Helix-Loop-Helix-Per-Arnt-Sim) family of transcriptional regulators. Located on chromosome 12q23.3, this gene produces a 593-amino acid protein that is expressed predominantly in the brain, where it plays critical roles in neural development, synaptic plasticity, cognitive function, and circadian rhythm regulation[@npas2020]. NPAS3 has emerged as a significant gene in both neurodevelopmental and neurodegenerative disorders. Heterozygous deletions and mutations are associated with intellectual disability, schizophrenia, and autism spectrum disorders. Additionally, NPAS3 expression is altered in [Alzheimer's disease](/diseases/alzheimers-disease) brains, where it may contribute to neuronal dysfunction and cognitive decline[@transcription2019][@bruel2021].
