PDE5A Gene

Overview

PDE5A (Phosphodiesterase 5A) encodes a cyclic nucleotide phosphodiesterase that specifically hydrolyzes cyclic guanosine monophosphate (cGMP). While classically studied in the context of smooth muscle relaxation and erectile dysfunction, PDE5A has emerged as an important regulator of neuronal function, synaptic plasticity, and neuroprotection in the central nervous system. Dysregulated PDE5A activity has been implicated in Alzheimer's disease, Parkinson's disease, and other neurodegenerative conditions[@rybalkin2019].

Molecular Biology

Gene Structure

The PDE5A gene spans approximately 50 kb on chromosome 4q27 and contains 26 exons. Three alternative splicing events produce multiple isoforms:

  • PDE5A1 — Full-length canonical isoform
  • PDE5A2 — Alternative N-terminus isoform
  • PDE5A3 — Testis-specific isoform

Protein Structure

PDE5A is a homodimeric enzyme with each monomer containing:
