Overview

The ROBO3 (Roundabout Guidance Receptor 3) gene encodes a transmembrane receptor protein that plays critical roles in axon guidance and neuronal development. Unlike other ROBO family members (ROBO1 and ROBO2) which primarily mediate repulsive axon guidance, ROBO3 has unique functions in the development of commissural neurons and is essential for proper formation of major neural pathways in the developing brain and spinal cord [1](https://pubmed.ncbi.nlm.nih.gov/10773876/). ROBO3 is particularly important for horizontal gaze control and midline crossing of axons, and mutations in this gene cause a rare neurodevelopmental disorder called Horizontal Gaze Palsy with Progressive Scoliosis (HGPPS) [2](https://pubmed.ncbi.nlm.nih.gov/20451879/).
