TFB1M (Transcription Factor B1, Mitochondrial) is a human gene located on chromosome 6p24.2 that encodes a mitochondrial matrix protein with dual enzymatic activities as both a transcription initiation factor and an rRNA methyltransferase. The methylation of 12S rRNA by TFB1M occurs co-translationally during ribosomal assembly. Loss of TFB1M methyltransferase activity results in defective small subunit assembly, leading to impaired mitochondrial translation. This deficiency manifests as a severe reduction in the synthesis of all 13 mitochondrial-encoded proteins, which are essential components of the OXPHOS complexes.
