Function
The UGCG gene encodes UDP-glucose ceramide glucosyltransferase (UGCG), also known as glucosylceramide synthase. This enzyme catalyzes the first step in the biosynthesis of glucosylceramide, a precursor for complex glycosphingolipids (GSLs). GSLs are abundant in neuronal membranes and play critical roles in neuronal development, signal transduction, synapse formation, and myelin maintenance.
In the brain, UGCG is essential for the formation of gangliosides, which are particularly enriched in the central nervous system. The enzyme is localized to the Golgi apparatus where it transfers glucose from UDP-glucose to ceramide. Dysregulation of glycosphingolipid metabolism has been implicated in several neurodegenerative diseases, including Parkinson's disease and related synucleinopathies.
Studies have shown that UGCG activity influences [alpha-synuclein](/proteins/alpha-synuclein) aggregation and toxicity. Inhibition of UGCG has been explored as a therapeutic strategy to reduce glycosphingolipid accumulation in lysosomal storage disorders and potentially modify disease progression in Parkinson's disease.
