Amyloid-Beta Metalloproteinases in Alzheimer's Disease

Overview and Background

[Alzheimer's disease](/diseases/alzheimers-disease) (AD) represents the most prevalent neurodegenerative disorder globally, affecting over 55 million individuals worldwide and imposing substantial socioeconomic burdens on healthcare systems [@alzheimers2023]. The disease is characterized by progressive cognitive decline, memory impairment, and accumulation of two hallmark pathological lesions in the brain: extracellular amyloid plaques composed of [amyloid-beta](/proteins/amyloid-beta) (Aβ) peptides and intracellular neurofibrillary tangles formed by [hyperphosphorylated tau](/proteins/tau-protein) protein [@selkoe2016]. Among these pathological features, the amyloid hypothesis has long dominated AD research, proposing that the accumulation and aggregation of Aβ peptides serve as the primary trigger for downstream neurotoxicity, synaptic dysfunction, and neuronal loss [@karran2011].
