Overview

Chronic Traumatic Encephalopathy (CTE) is a progressive neurodegenerative disease associated with repetitive traumatic brain injury (TBI), including concussive and subconcussive blows commonly sustained in contact sports such as American football, boxing, hockey, and soccer, as well as military combat veterans exposed to blast injuries.[@mckee2013] First described in 1928 as "dementia pugilistica" in boxers, CTE is now recognized as a distinct tauopathy with unique pathological features and clinical manifestations that can develop years or decades after the initial head trauma. [@mez2017]

CTE is characterized by the abnormal accumulation of hyperphosphorylated tau protein (p-tau) in neurons, astrocytes, and around blood vessels, forming neurofibrillary tangles (NFTs) and neurites in a pattern distinct from other neurodegenerative diseases.[@mckee2013] The disease typically progresses through four pathological stages, correlating with increasing clinical severity.
