Evidence Summary: BECN1 is the master regulator of autophagy initiation, forming the core PI3K-III complex. BECN1 protein levels are reduced 30-50% in AD and PD brain tissue["@pickford2008"]. BECN1+/- mice spontaneously develop neurodegeneration, amyloid and tau pathology, and motor/behavioral deficits["@lucaszhang2014"]. AAV-BECN1 delivery reduces amyloid plaques by ~50% and protects dopaminergic neurons in PD models["@spencer2009"]. BECN1 occupies the most upstream position in the autophagy cascade — restoring it would benefit all downstream autophagy-dependent processes.
