mTOR Signaling Pathway in Neurodegeneration

Overview

The mammalian target of rapamycin (mTOR) signaling pathway is a central regulator of cell growth, metabolism, protein synthesis, and autophagy. [mTOR](/mechanisms/mtor-signaling-pathway-pathway) integrates signals from nutrients, growth factors, energy status, and stress to coordinate cellular homeostasis. In neurodegenerative diseases, [mTOR](/mechanisms/mtor-signaling-pathway) signaling is frequently dysregulated, contributing to impaired autophagy, abnormal protein aggregation, synaptic dysfunction, and neuronal death. The mTOR pathway represents a promising therapeutic target, with mTOR inhibitors like rapamycin showing neuroprotective effects in preclinical models of Alzheimer's disease (AD), Parkinson's disease (PD), and other neurodegenerative disorders.
