NRF2 Signaling in Oxidative Stress and Neurodegeneration

Overview

NRF2 (Nuclear factor erythroid 2-related factor 2), encoded by the [NFE2L2](/genes/nfe2l2) gene, stands as the master regulator of cellular antioxidant response, controlling over 500 target genes involved in oxidative stress defense, detoxification, mitochondrial function, and inflammation[@nrf2015] PMID: 32321531. First characterized in 1995 as a transcriptional activator of the β-globin gene, NRF2 has emerged as a critical protector against oxidative damage in virtually every tissue, including the brain[@nrf1995] PMID: 39962463. Dysregulated NRF2 signaling contributes to pathogenesis in [Alzheimer's disease (AD](/diseases/alzheimers-disease))[@nrf2015a], [Parkinson's disease (PD](/diseases/parkinsons-disease))[@nrf2013], [amyotrophic lateral sclerosis (ALS](/diseases/amyotrophic-lateral-sclerosis))[@nrf2012], [Huntington's disease (HD](diseases/huntingtons))[@nrf2015b], [multiple sclerosis (MS](/diseases/multiple-sclerosis))[@nrf2015c], and increasingly recognized in frontotemporal dementia and prion diseases PMID: 40663116.
