Oxidative Stress in Neurodegeneration

Introduction

Oxidative stress represents one of the most fundamental and early pathogenic mechanisms in neurodegenerative diseases, including [Alzheimer's disease](/diseases/alzheimers-disease) (AD), [Parkinson's disease](/diseases/parkinsons-disease) (PD), Amyotrophic Lateral Sclerosis (ALS), and Huntington's disease (HD)[@butterfield2023][@dias2023]. Defined as an imbalance between the production of [reactive oxygen species](/entities/reactive-oxygen-species) (ROS) and the cellular antioxidant defense capacity, oxidative stress contributes to neuronal dysfunction and death through multiple pathways, including lipid peroxidation, protein oxidation, DNA damage, and mitochondrial dysfunction[@prez2024][@fischer2023]. The brain is particularly vulnerable to oxidative damage due to its high metabolic rate, elevated oxygen consumption, and relatively limited antioxidant capacity compared to other organs[@cobley2023].
