Introduction

[Synapse loss](/mechanisms/synapse-loss-mechanisms) represents one of the earliest and most robust pathological features of [neurodegenerative diseases](/mechanisms/neurodegeneration-pathways), correlating directly with cognitive decline in [Alzheimer's disease](/diseases/alzheimers-disease), [Parkinson's disease](/diseases/parkinsons-disease), and related disorders. The elimination of synapses precedes [neuronal death](/mechanisms/neuronal-loss-apoptosis) and occurs decades before clinical symptoms manifest in many cases.
