TREM2 Signaling in Neurodegeneration

Introduction

Triggering Receptor Expressed on Myeloid Cells 2 (TREM2) is a critical immune receptor expressed primarily on microglia in the central nervous system (CNS) that plays a pivotal role in regulating neuroinflammatory responses, microglial survival, and phagocytic function. Since the discovery that loss-of-function variants in the TREM2 gene significantly increase the risk of Alzheimer's disease (AD) approximately three- to fourfold, TREM2 has emerged as a central player in neurodegenerative disease pathogenesis [1](https://doi.org/10.1016/j.neuron.2014.05.004). This page provides a comprehensive analysis of TREM2 structure, signaling mechanisms, downstream molecular pathways, and therapeutic implications for neurodegenerative diseases. [@guerreiro2013a]
