YAP/TAZ (Hippo) Signaling in Neurodegeneration

Overview

The Hippo signaling pathway is a highly conserved kinase cascade that controls organ size by regulating cell proliferation, [apoptosis](/entities/apoptosis), and stem cell renewal. In the central nervous system, YAP (Yes-associated protein) and TAZ (transcriptional coactivator with PDZ-binding motif) have emerged as critical regulators of neuronal survival, neurogenesis, and neurodegeneration. Dysregulation of Hippo pathway signaling has been implicated in Alzheimer's disease, Parkinson's disease, and other neurodegenerative disorders.

Pathway Components

Core Kinase Cascade

The Hippo pathway consists of a kinase cascade that ultimately controls YAP/TAZ activity:

  • MST1/2 (hippo): Mammalian STE20-like kinases 1 and 2, the upstream kinases[@mst]
