Nicastrin (NCSTN) is a type I transmembrane glycoprotein that serves as an essential cofactor for the gamma-secretase complex. It plays a critical role in the assembly, stability, and catalytic activity of gamma-secretase, which is responsible for the intramembranous cleavage of [amyloid precursor protein](/entities/app-protein) (APP) and other type I transmembrane substrates.
Structure
Nicastrin is a large single-pass transmembrane protein with complex domain organization.
