Structure

Prosaposins are precursor proteins containing multiple functional domains:

• Signal peptide: N-terminal targeting sequence
• Saposins A, B, C, D: Four homologous domains (~80 aa each)
• Each saposin is generated by proteolytic cleavage

• Compact, globular proteins
• Six conserved cysteine residues forming three disulfide bonds
• Form saposin-like pores in membranes

Normal Function

Prosaposin is a lysosomal precursor protein that is processed into four saposins (A, B, C, D), each with distinct functions:

Saposins:

• Saposin A: Activates glucocerebrosidase (hydrolyzes glucosylceramide)
• Saposin B: Activates arylsulfatase A (hydrolyzes cerebroside sulfate)
• Saposin C: Activates β-glucosidase and β-galactocerebrosidase
• Saposin D: Activates ceramidase
