Overview
SGK1 (Serum/Glucocorticoid-Regulated Kinase 1) is a serine/threonine protein kinase belonging to the AGC family. Originally identified as a glucocorticoid-responsive gene, SGK1 is a critical regulator of cell survival, metabolism, and stress responses in the nervous system [@supsup2022]. SGK1 is ubiquitously expressed with particularly high levels in the brain, including the [hippocampus](/brain-regions/hippocampus), [cortex](/brain-regions/cortex), and basal ganglia [@supsup2021].
Structure
SGK1 possesses the characteristic architecture of AGC kinases:
- N-terminal kinase domain (~280 residues): Contains the catalytic site with activation loop (Thr256)
- Akt-like hydrophobic motif (FXXFS/TY): Regulatory site (Ser422) phosphorylated by mTORC2
- PDK1 phosphorylation site: Thr256 in the activation loop
- N-terminal variable region: Contains nuclear localization signals
