Overview

SMAD Family Member 5 (SMAD5) is a critical mediator of bone morphogenetic protein (BMP) signaling, playing essential roles in embryonic development, neurogenesis, synaptic plasticity, and cellular homeostasis. As a receptor-regulated SMAD (R-SMAD), SMAD5 transduces extracellular BMP signals from the cell surface to the nucleus, regulating gene expression programs that control neural stem cell fate, neuronal differentiation, and glial cell development.[@wang2019] Recent research has revealed important roles for SMAD5 in neurodegenerative disease pathogenesis, making it a potential therapeutic target for conditions including Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS).[@liu2018]
