## TRPV1 Protein is a protein. This page describes its structure, normal nervous system function, role in neurodegenerative disease, and potential as a therapeutic target.
TRPV1 Protein
Structure
TRPV1 (Transient Receptor Potential Vanilloid 1) is a non-selective cation channel belonging to the TRP (Transient Receptor Potential) channel superfamily. The channel is a tetramer composed of four identical subunits, each containing six transmembrane domains (S1-S6) with a pore-forming loop between S5 and S6[@nilius2014]. The N-terminus contains ankyrin repeat domains (ARD) that are involved in protein-protein interactions and channel regulation[@venkatachalam2007].
Key structural features:
- Ankyrin Repeat Domain (ARD): Six ankyrin repeats in the N-terminus for protein interactions
- S1-S4 domain: Voltage-sensor-like domain that responds to ligands
- S5-S6 pore domain: Forms the ion conduction pathway
- Vanilloid binding pocket: Hydrophobic pocket for capsaicin and resiniferatoxin binding
Normal Function in the Nervous System
TRPV1 is expressed in sensory [neurons](/entities/neurons) (dorsal root ganglion, trigeminal ganglion), where it functions as a polymodal nociceptor activated by[@caterina2000]:
- Thermal stimuli: Noxious heat (>43°C)
- Chemical stimuli: Capsaicin (from chili peppers), resiniferatoxin, protons (acidic pH)
- Mechanical stimuli: Moderate mechanical force
In the central nervous system, TRPV1 is expressed in:

Physiological functions include:

- [Amyloid-beta](/proteins/amyloid-beta) interaction: Aβ oligomers sensitize TRPV1, leading to increased calcium influx and neuronal dysfunction
