Myelin Repair Therapies represent a promising therapeutic strategy for neurodegenerative diseases characterized by demyelination and white matter pathology, including multiple sclerosis (MS), Alzheimer's disease (AD), Parkinson's disease (PD), and vascular dementia[@rodriguez2014]. - Myelin production: Synthesize and maintain the multilamellar myelin membrane

- Axonal stability: Produce neurotrophic factors that support axonal survival
Demyelination in Neurodegenerative Disease
While traditionally associated with multiple sclerosis, demyelination and oligodendrocyte dysfunction contribute to pathology in several neurodegenerative conditions:

Oligodendrocyte Precursor Cell (OPC) Activation
OPCs are present throughout the adult CNS and can differentiate into mature oligodendrocytes to remyelinate axons. Therapeutic strategies include:

Conclusion

While MS has provided proof-of-concept, translation to AD, PD, and other conditions requires understanding disease-specific mechanisms of oligodendrocyte dysfunction. The combination of pharmacological, cell-based, and supportive approaches may prove most effective. Ongoing clinical trials will clarify the potential for myelin repair strategies in non-MS neurodegeneration.
