Antisense Oligonucleotide Chemistry
The chemical backbone of ASOs significantly impacts their pharmacological properties. Modern ASOs employ several key modifications:

RNase H-Dependent Mechanism

When the gapmer binds to target mRNA, the DNA:RNA hybrid recruits RNase H, which cleaves the RNA strand while leaving the ASO intact for additional catalytic cycles.

Splicing Modulation
