While motor neurons release mtDNA cell-autonomously, astrocytes phagocytose dying neurons and encounter released mtDNA, activating astrocyte cGAS/STING to induce chronic inflammatory phenotype (CXCL10, IL-6, complement). However, oligodendrocyte-derived IL-6 may prime microglia through IL-6 trans-signaling (IL-6/sIL-6R/gp130), creating a self-reinforcing inflammatory loop.
