Gain-of-function experiments were performed to study the role of 3-hydroxy-3-methylglutaryl-CoA synthase 2 (HMGCS2) in regulating alveolar epithelial cell lipid metabolism. Ectopic expression of HMGCS2 in AECIIs was achieved to investigate its effects on lipid degradation and cellular function. The experiments demonstrated that HMGCS2 promotes lipid degradation through interaction with PPARα and upregulation of CPT1A and CPT2 expression.
PRIMARY OUTCOME
lipid metabolism restoration
EXPECTED OUTCOMES
Enhanced lipid degradation and improved AECII function
SUCCESS CRITERIA
Increased CPT1A/CPT2 expression and reduced lipid accumulation
PROTOCOL
Overexpression of HMGCS2, assessment of lipid degradation markers and CPT1A/CPT2 expression
