This in vitro experiment tested whether LPT1 could protect cultured hepatocytes against cell death induced by pro-PANoptosis molecules under lipid stress conditions. Hepatocytes were treated with LPT1 (100 nM) and then challenged with PANoptosis-inducing agents including TNF-α, LPS, and nigericin in the presence of lipid stress to create an in vitro MAFLD model. The experiment aimed to validate the protective effects of LPT1 observed in the mouse model and specifically test its ability to prevent PANoptosis-mediated cell death in hepatocytes. This complemented the in vivo findings by providing mechanistic insights into how LPT1 protects against the coordinated cell death pathway involving apoptosis, pyroptosis, and necroptosis.
