Flow cytometry and western blot analysis were performed on blood and brain samples from mice overexpressing human α-synuclein to assess toll-like receptor 2 and 4 expression. This experiment complemented the human studies by examining whether α-synuclein overexpression directly affects TLR expression in an animal model. The study found no significant changes in TLR2 and TLR4 expression in blood and brain of these transgenic mice at 12 months of age, despite the expected α-synuclein pathology. This suggests that the TLR changes observed in human patients may be due to additional factors beyond α-synuclein overexpression alone.
