🧫 Experiment Protocol
Exploratorymetabolic syndromeRUBCNPTEC-hepatocyte transwell co-cultureproposed
Performed transwell co-culture experiments with RUBCN knockout PTECs and hepatocytes (BNL-CL2 cells) to investigate fatty acid efflux. Found that KO PTECs promoted massive triglyceride accumulation in co-cultured hepatocytes, suggesting accelerated fatty acid efflux from the PTECs. This mechanism was proposed to contribute to the metabolic syndrome observed in KO mice.
PRIMARY OUTCOME
triglyceride accumulation in hepatocytes
EXPECTED OUTCOMES
Increased fatty acid efflux from KO PTECs leading to hepatocyte triglyceride accumulation
SUCCESS CRITERIA
Quantification of triglyceride levels in hepatocytes
PROTOCOL
Co-cultured RUBCN KO PTECs with BNL-CL2 hepatocytes in transwell system, measured triglyceride accumulation
