Validation experiment designed to validate causal mechanisms targeting NR1D1,NR1D2 in C57BL/6J mice. Primary outcome: Behavioral antidepressant response to sleep deprivation
This experiment tested whether pharmacological activation of the circadian clock repressor REV-ERB would suppress the antidepressant effects of sleep deprivation. The study demonstrated that REV-ERB agonist treatment blocked the rapid antidepressant behavioral effects normally produced by sleep deprivation, providing pharmacological evidence for the role of circadian clock mechanisms in mediating SD's therapeutic effects.
REV-ERB agonist administration followed by sleep deprivation and behavioral assessment
Suppression of sleep deprivation's antidepressant effects by REV-ERB activation
Significant reduction in antidepressant behavioral response compared to vehicle-treated controls
