This experiment investigated the role of connexin 43 (Cx43) in mediating mitochondrial transfer by knocking down Cx43 expression in MSCs using siRNA. After knockdown, MSCs were co-cultured with chondrocytes and mitochondrial transfer was quantified. The study found that knocking down Cx43 significantly decreased mitochondrial transfer from MSCs to chondrocytes, demonstrating that Cx43 expression is necessary for efficient mitochondrial transfer. This experiment provided evidence that Cx43 is a key mediator of the mitochondrial transfer process.
