Exploratory experiment designed to discover new patterns targeting NPM1 in OVA-presenting tumor cells and OT-1 transgenic T cells. Primary outcome: T-cell mediated tumor cell killing efficiency
Assessment of antigen-specific T-cell mediated killing of tumor cells presenting ovalbumin (OVA) by OT-1 transgenic T cells. This experiment evaluated how NPM1 deficiency affects the ability of tumor cells to present antigens and be recognized and killed by specific cytotoxic T lymphocytes. The study demonstrated that Npm1 deficiency increased MHC-I and MHC-II molecule expression and enhanced specific T-cell killing capacity.
In vitro co-culture of OVA-presenting tumor cells with OT-1 transgenic T cells to measure antigen-specific cytotoxicity
NPM1 deficiency would enhance T-cell recognition and killing of tumor cells
Increased cytotoxic T lymphocyte-mediated tumor cell death
