Flow cytometry and western blot analysis were performed to detect toll-like receptor 2 and 4 expression in blood and brain samples from Parkinson's disease patients. The study examined circulating monocytes and B cells in blood samples, as well as brain tissue from the caudate/putamen region. Results showed increased TLR2 and TLR4 expression in circulating monocytes, increased TLR4 in B cells, and increased TLR4 in the caudate/putamen of Parkinson's disease patients compared to controls. This clinical study provides evidence for immune system activation and toll-like receptor upregulation in Parkinson's disease patients, supporting the hypothesis that inflammation contributes to disease pathogenesis.
