Independent replication study using the Generation Scotland: Scottish Family Health Study (GS:SFHS) cohort to validate genetic variants identified in the initial meta-analysis. A total of 171 SNPs with P < 1 × 10^-7 from the meta-analysis were tested for replication in 2,659 MDD cases diagnosed with DSM-IV criteria and 17,237 controls. The replication analysis identified one additional genome-wide significant locus and confirmed the association of previously identified variants.
PRIMARY OUTCOME
replication of genetic associations with MDD
EXPECTED OUTCOMES
Validation of genetic associations identified in discovery meta-analysis
SUCCESS CRITERIA
P < 5 × 10^-8 in combined analysis
PROTOCOL
Genotyping and association testing of 171 prioritized SNPs in independent Scottish cohort. Cases diagnosed using DSM-IV criteria. Combined analysis with discovery cohorts.
