🧫 Experiment Protocol Validationdiet-induced cognitive dysfunctionCHRNA7adult Swiss male miceproposed
Adult Swiss male mice were fed either standard chow or 60% high-fat diet for 3 days to evaluate the effects of brief HFD exposure on hippocampal cholinergic function and cognitive behavior. Mice underwent behavioral testing using Open Field and Novel Object Recognition tests to assess anxiety-like behavior and memory performance. The CA3 region of the hippocampus was then analyzed using Western Blotting, PCR, and immunofluorescence to examine α7 nicotinic acetylcholine receptor expression, acetylcholinesterase levels, and neuroinflammation markers. This experiment aimed to establish the relationship between short-term high-fat diet consumption and cholinergic dysfunction in the hippocampus.
PRIMARY OUTCOME
memory performance on Novel Object Recognition test
EXPECTED OUTCOMES
HFD would impair memory performance and reduce α7nAChR expression while increasing neuroinflammation
SUCCESS CRITERIA
Significant differences in NOR performance and molecular markers between HFD and control groups
PROTOCOL
3-day feeding of 60% HFD vs standard chow, followed by Open Field and Novel Object Recognition behavioral tests, then tissue analysis of CA3 hippocampal region using Western Blotting, PCR, and immunofluorescence
