RAW264.7 macrophages were treated with oxidized low-density lipoprotein (ox-LDL) to simulate atherosclerotic conditions. Quantitative real-time PCR (qRT-PCR) was performed to confirm the expression levels of hub genes C1QA, C1QC, and the transcription factor SPI1. This experiment validated the computational findings from the transcriptomic analysis by demonstrating that these genes are indeed upregulated in macrophages exposed to ox-LDL, which is a key trigger of atherosclerotic inflammation.
