🧫 Experiment Protocol
ExploratoryCD9TIGER mice cortical astrocytes and microgliaproposed
Cortical astrocytes from TIGER mice were shown to release approximately 136 nm extracellular vesicles containing CD9. These EVs were injected intraventricularly and their uptake was studied. CD11b/IBA1 positive microglia surrounding the lateral ventricles were found to take up the injected EVs, demonstrating the transfer of astrocyte-derived EVs to microglia in the brain. This experiment provided evidence for intercellular communication via EVs between astrocytes and microglia in the central nervous system.
PRIMARY OUTCOME
EV uptake by microglia following intraventricular injection
EXPECTED OUTCOMES
Microglia uptake of astrocyte-derived CD9-positive EVs
SUCCESS CRITERIA
Detection of CD9-GFP positive EVs in CD11b/IBA1 positive microglia
PROTOCOL
EV isolation from astrocytes, size characterization, intraventricular injection, immunofluorescence analysis of microglia uptake
